On human DNA, it would not have worked because the oligonucleotide would not have specifically bound to a single site. On a DNA as complex as human DNA it would have bound to hundreds or thousands of sites depending on the sequence involved and the conditions used. What I needed to make this work was some method of raising the relative concentration of the specific site of interest. What I needed was PCR, but I had not considered that possibility. I knew the difference numerically between five thousand base pairs as in a plasmid and three billion base pairs as in the human genome, but somehow it didn't strike me as sharply as it should have. My ignorance served me well. I kept on thinking about my experiment without realizing that it would never work. And it turned into PCR.
I have soaked 1 or 2 cups of poppy seeds in lemon juice for about an hour; the acidic juice eats through the skin of the seed and releases the opium. I then strained the juice into a cup, added cold water and heaps of honey, and ice. I can attest the resulting drink is more potent than many of the prescription pill opiates. My flatmate was studying to be a pharmacologist so I felt comfortable with his assurance that I wouldn’t die, but I could see it becoming as addictive as anything else. So yes, you can get a dose from just seeds. Carefully.
There are 10 sections in all in the test. The first section is writing essay (25 minutes), second is Critical Reading which includes Sentence Completion, Short Reading Passages, Long Reading Passages. Duration is 25 minutes. Next section is Mathematics (25 minutes). The fourth section is Writing- Multiple Choice and the fifth section is Critical Reading which includes Sentence Completion, Short Reading Passages, Long Reading Passages. Time allotted for both the sections is 25 minutes each. The sixth section is again Mathematics, multiple choice questions (25 minutes). The seventh section is again critical reading and next is Mathematics. Both of these are of 20 minutes duration. Next section is critical reading of 25 minutes and the last section is Writing- Multiple Choice which is of 10 minutes duration and includes Improving Sentences.
